Obtain six petri dishes containing synthetic "dropout" medium "SD-trp"
|
|
label one petri dish as no fabric and the rest with either cotton, polyester, linen, nylon, or rayon
|
|
Place the six petri plates on the bench right side up
|
|
obtain an original yeast suspension with cells that bears the allele trp1-289
|
|
pipette 100 micro liters of the suspension onto each petri dish
|
|
|
|
|
|
|
|
|
|
replace cover of each dish
|
|
wait a few seconds for the spreader to cool, then carefully spread the suspension over the entire plate
|
|
remove the glass spreader from the alcohol and pass it through the flame
|
|
place a glass spreader in a beaker containing 95% alcohol and light a flame
|
|
replace the cover of each dish after pipetting
|
|
|
|
|
|
|
|
|
|
make a foil cap for the UV irradiation lamp and turn on the short wave UV lamp and cover with cap
|
|
cover each plate with the fabric labelled on the dish. The no fabric dish should only have saran wrap
|
|
doing two at a time: place the dishes under the lamp and remove the cap
|
|
allow the petri dishes to sit under the lamp for the number of seconds found in the previous lab that induces mutations.
|
|
replace cover for each dish and tape all 6 together upside down. allow plates to be induced at 30 degrees for 2 days & then placed in a refrigerator
|
|
|
|
|
|
|
|
|
